Gilgamesh. But that's the smartass answer, because he wasn't in a comic book. Nevertheless, he was a super hero: he was super-powerful, he fought super-enemies and he had dramatic adventures. He even had a sidekick who died, Enkidu.

But the first comic book superhero of the kind we call superhero was Superman. There were other costumed comic book heroes that preceded him, but they are usually known as 'mystery men', like the Shadow (1930), Mandrake (1934), Doctor Occult (1935), the Phantom (1936), that sort of thing.

Actually it was Mandrake the Magician. He first appeared in 1934, predating Superman by 4 years.
