"make" means to cause something to happen. "make up" means to constitute, to come together as a whole.

The day was dull. Then we received a package in the mail from my aunt. Getting that package made the day much better (caused it to be).

Each moment of the day was unexciting. Those were the moments that made up a dull day (came together as consistently dull so that the whole day was dull).

The 50 individual states make up the US as a whole.

The founders made the US by declaring it an independent country.

Several books make up the Harry Potter series.

J.K. Rowling made the Harry Potter books.

The important part is the ticking.

Ticking away the moments that make up a dull day

Fritter and waste the hours in an offhand way.

    There is no difference .
