    You should call to confirm that your letter or email has been received, and politely ask if everything is all right and that you are at their disposal if they need any more info.

    I would first send an e-mail as hard copies of your application may get lost if you hand it in.

    Then if you don't hear anything after 3/4 days, you should politely call and ask the current status of your application and state how much you want to work there.
