Which Melbourne Suburbs would you recommend for a professional couple?
We are looking for somewhere which is close to the CBD which has good transport connections and is reasonable to rent / buy?

St Kilda, Prahran,Sth Yarra,Albert Park are all great areas close to the CBD with great public transport facilities. However these suburbs tend to be quite pricey.
Maybe try Carlton or Fitzroy or maybe even Richmond

Carlton is great for a professional couple - if you head a bit more north in the inner city suburbs it's alot more reasonable.
North Fitzroy which has awesome cafes - Brunswick East/Northcote is quite cheap but with nice houses and not too nosy plus loads more trams and better atmostphere then the south side suburbs like prahran or south yarra.

Bentleigh, Bentleigh East and Oakleigh are all pretty close to the CBD from the Monash Fwy (or carpark..?) and will be pretty affordable. In the other directions; Nunawading, Mitcham and Mt Waverly are all close to the Eastern Fwy, and so, not far from the CBD but traffic is real bad. Templestowe is also a really good area, with close access to the Eatstern.

Prices are so high these days you won't find anything for 300K!!! But I think Clayton is still regarded as being fairly cheap compared to surrounding suburbs. Maybe you can get something for 400-450K

Depends on what you want.
You can have the multicultural Carlton, Fitzroy, Northcote or you can have the upmarket (snobby) area like Brighton.
For travel to the City Centre, i'd go with Carlton, Northcote or Fitzroy, easy & as close as you can get.

3km from the city and about 30% cheaper than Carlton......

Carlton, it is fairly upper class, but has great cafes/shops/fashion boutiques.
Not to mention THRESHERMANS!
Best bakery ever...coffee guy who's always walking there, rules.
