Really depends on your tastes you can barely realy see everything in 2 museums in one day and there like 10 of them down at the Mall. + the zoo and other stuff.
The archives have the constitution and bill of rights you cant see those anywhere else, washingon memorial is cool but most time you gotta wait in line and that can kill an hour, lincoln is a must but it is all the way at the end of the mall so do it 1st or last, if you can get into the city butt early you can try to score tickets to the white house tour, spy museum is something different but you do have to pay for it and its not located at the mall but you will be in chinatown and behind the sports arena there are some athentic chinese rest. for lunch (you know their athentic cause they have meet hanging on hooks in the front window:) the zoo is great if the weather is nice but this will take up most of a day if you want to see all the animals. Ride into the city on metro so you dont have to worry about traffic or parking. Air and space is probably the most popular with the kids if you have any. If you are going to be in baltimore with your friend the aquarium up there is pretty cool also.

That's a lot to try to do in one day. Depending on how long you want to spend in each place, I would cut that list down by maybe half. Different things take different amounts of time, though; for example, the Iwo Jima Memorial would probably only take a few minutes to visit, whereas the White House or Capital could take over an hour each. Similarly, you could spend hours in a Smithsonian museum, although you could also just make a quick visit. My suggestion is to pick the things from that list that you want to do most, and do those first as you might not have time for much else. Also, the Smithsonian, Capitol, and many memorials are in the same area near the National Mall, so you could do all those things at once.

We just did a podcast on what we called the "Grand Tour" of Washington, walking around the Capitol, up Pennsylvania Avenue to the White House then across to the Washington Monument, and then to the Tidal Basin. Takes about 2 hours if you don't stop anywhere.
But you could do this then walk up the mall to the Lincoln Memorial.
All that said, your best bet might be Tourmobile - a bus that, for 24 dollars or so gives you on and off priveliges for the whole day. It runs along the mall from Union Station to the Lincoln Memorial and will save much in the way of tired feet.
There are about 10 Smithsonian museums on the mall, they are all good, pick one based on your interests, and spend a couple of hours in it.
If you want to visit the inside of the Capitol, then get in line early. For the Washington Monument you can get tickets in advance and save time waiting in line.

Go online to Washington DC tours and pick a site. I like the Onboard Tour that lasts 6 hours, but really gives a nice overview of the city and its many, diverse sites. It costs $60 per adult, but is well worth the price. You'll need a reservation because......
We're in the middle of the Cherry Blossom Festival right now, so expect lots of crowds. And be sure to bring a jacket or warm sweater. Temps can be pretty cold at this time of year (mid-40's) and nights frosty.

It all depends on what you are interested in...
If you like American History then all the monuments and the American History Museum is great; also the Air and Space Museum is great. If you want to see the White House or go up in the Washington Monument then you need to go and get tickets early as they go fast.
There is also the Natural History Museum which a lot of people enjoy; also the National Art Gallery is very good with everything from Textile art to Modern Art.
I would also recommend buying one of the tour bus tickets that saves a lot of walking time also some of them go out to Crystal Lee and Arlington Cemetery if you just stay on the bus you get to see a lot of stuff and are told lots of interesting things.
If you like Aquariums then I would suggest going up to Baltimore.
