1 teaspoon or 5mL of children's benadryl active ingredient diphenhydramine 12.5mg
A very conservative dose would be 2.5mL.
My son began taking the 5mL at 18 months old per doctors orders, however he is larger than most, at two he weighed 30 lbs and is 36" now at 2.5yrs

this conservative site which is a peds office lists the dose as 2.5ml if you haven't ever used start with that but that in my opinion is way too low of a dose to be effective

24-35 lbs is considered a 2 year old
My son has been on benadryl since before he turned 1 per doctors orders (he has itchy eczema)

My pediatrician recommended it to help during teething. 2.5 ml was the dose we gave and still give my 17 month old and my 3 year old... they have a 4 lb weight difference.

I really don't think you should be giving a 2 year old benadryl at all. That stuff is hardcore. It knocks ME out. I'm pretty sure your toddler would be sleeping for a week.

In infants and children, especially, antihistamines in overdosage may cause hallucinations, convulsions, or death. As in adults, antihistamines may diminish mental alertness in children. In the young child, particularly, they may produce excitation.
