What is the most significant event or time in human history?
It could be in your lifetime or not. 9/11...MLK's march on Washington...JFK being assassinated...Columbus sailing to the new world...fire being used by cavemen...anything. And why? :)

Besides the birth and death of Christ, the Enlightenment was because it not only influenced European and Asian history, but American history as well.
The Enlightenment was an 18th-century intellectual movement in Western Europe that emphasized reason and science in philosophy and in the study of human culture and the natural world. This led to the Darwinian Revolution (which influenced man's belief systems), the World Wars (which was, although a horrific time of bloodshed and death, a time of progress and technological develpment), and the Cold War which was the time of containment and isolation of communism.
And from there was the rest of history until now. You can see why the Enlightenment was a very influential time in history for humanity.

Columbus' discovery of the Western Hemisphere is the most significant event in human history. Because for the first time in recorded history the two worlds were united despite the difficulties in reaching either side. Yes, I know the Vikings lay a fair claim to the discovery and Columbus may not have made the voyage had it not been for Viking knowledge. What Columbus did different was he sailed a far more southern route that could not be confused as just another ice-free area that included Greenland and the Artic Ocean.
New maps of the entire world had to be made, new words were made, used, and incorporated into Eastern Hemisphere languages, new foods, raw material. Better means of oceangoing navigation evolved and new ideals were given a better chance than earlier Eastern Hemisphere history normally allowed. More lives were changed by Columbus' discoveries than any other event in world history on both side of the Atlantic and even in Asia; because it was spared invasion of white Europeans until a later date.

I'd say it was the Greek victory at Salamis, to choose a single point of divergence that is known well.
