It is about a 2 and a half hour drive from Seattle to Vancouver.
If you can, it is better to spend the night in Vancouver and check out the night life there.
I once did Seattle-Vancouver-Seattle in a day, but I already knew Vancouver and was taking a tour with some friends. I left very early in the morning and returned in the evening.
The border crossing is not so bad....around 15 minutes wait.

You need a passport these days. The drive is NOT 5 hours - it's 2 1/2 to 3 hours depending on day of the week, etc. You will be charged a VAT tax for anything you buy in Canada, but there is a form you can submit for a refund. Keep receipts for that and also to verify your border crossing statement (if required) upon return. Be sure to check the exchange rate ($) before you go so you don't get ripped off if you pay cash. Be sure to see Stanley Park and Granville Island. You'll drive past Burnaby on the way - Michael J. Fox's hometown. Oh, and if you have time, LaConner, Wa. is a neat little detour - especially during tulip/ daffodil time.

I live between the two cities and travel to both often.
It is about 140 miles between the two cities. I would estimate driving time to be about 3 hours one-way, including the average border wait. The border crossing is normally about 15 minutes going north and a bit longer going south. Although that can vary greatly. On a Saturday afternoon you might wait up to 2 hours to go south.
Do bring your passport and some patience.

Avoid rush hours at the border and your drive will be well under 3 hours. Vancouver has a lot of food and beaches. Passport required for non-border state residents.

It should take you about 5 hours. This is including time to cross the border. Bring your passport and another form of I.D. I would recommend staying at least one night though, Vancouver is a great place.

Despite the three hour drive, there is no way to predict how long the border crossing will take.
