"Why" is the question that needs to be attached to any "big" question.
To address the ideas from one of the other answers:
1) WHY did life start? (more important than how)
2) WHY does life have purpose?
3) WHY should we think about what comes after death?
4) WHY should we be concerned with right vs. wrong?
5) WHY?
Best wishes and good luck.

a lot of the philosophical 'big questions' surround our origin and destination with what is important in the middle. I believe in essence though the really important one is were we are at the moment of now, we could look at where one believes humanity will end and what is in store after that but the nature of philosophy means that these questions would change whith every answer we get.

What is or is there a Creator (God)? What is the Human Soul? What is the meaning of life ?(though I happen to know the answer to that one). What happens after we die? What is the Unified Theory of Relativity? Is there life on other worlds? What is the cure for all disease or ailments? What does a woman really want? And of course, is there really a Bigfoot?

I think that is the most valuable question I've ever read.
VALUE
What is to be valued?
How best to create value?
How best to alter our natures to fully experience the value all around us?
NATURE
How best to come to understanding the nature of the grandest thing observable, smallest thing observable?
How best to discover the nature of self?
How best to avoid becoming a victim of our own natures?
STRUCTURE
What morals & rules allow for the natural progression of all that is truly valuable to proliferate in society?
APPLICATION
How best to aid the process of adaptation in all that we do?
How best to apply what we know in our day to day lives?
This is a ridiculous attempt to sum up all of my questions into one big deep powerful question,
What structures allow for maximization of most valuable use of the limits of our capabilities with the extent of our resources for all parties, considering the natures therof in ways applicable to our day to day lives? Lol. I'm retarted. No man should ever lead others until he answers this question with absolute precision. Hahaha
