You're referring to a war time song which said "Hitler, has only got one ball". 

There's no evidence to support this (or technically refute it).  I don't think there was any publicly discovered medical records about Hitler which would have included this kind of information.  There has been long standing theorizing about the nature of Hitler's sexuality and sexual experience.  In an unusual move for a world leader of his era, Hitler never married (until a few days before his death).  He was also never officially linked with anyone romantically.  Hitler wanted to portray himself as a selfless leader, devoted to the nation.  So he eschewed any public romantic connection.  But whether or not there were other reasons for this isn't known.  Various people have speculated that Hitler's lack of public interest in women may have been because he was gay, or asexual, or had a more deviant sexual proclivity.  There's also been speculation that maybe he had a medical condition which made sex difficult or impossible, such as a venereal disease.  But we just don't know. 

The song itself, even if the accusation turns out to be true, is pure speculation.  They didn't know anything about Hitler's reproductive health.  They were just disparaging his masculinity by criticizing his anatomy
